First and foremost, I feel that Iago understands exactly what each individual requires in order for that character to fall for his falsehoods and trust him. Iago analyzes these personalities from head to toe, learning their strengths and flaws and exploiting them to ensure that his evil plans always succeed. So, with that in mind, Iago goes after Othello in order to persuade him to accept his falsehoods about Desdemona. The explanation for this is unclear, however I saw two possibilities. One motive might be because Othello promoted Cassio instead of Iago, or it could be that Iago believes Othello slept with his wife.
