Normally, all proteases are synthesized in the inactive state. Further, after isolation, they are activated by partial proteolysis (this is typical for most proteases). But in some diseases(most often with inflammation of the glands such as pancreas) proteases can be activated inside the cells then they begin to work already inside the cells and destroy it, and are immediately active
