In chemical science spectrophotometry is the quantitative measurement of the reflection or transmission properties of a material as a function of wavelength.
Firstly UV spectrophotometer is calibrated by a range of artificially prepared solutions of a known concentration. The obtained responses are used to draw a graph of concentration-response dependence.
After that a solution with an unknown concentration of a component is tested on the UV spectrophotometer. The obtained response is than compared with the previously produced graph and the component concentration is recovered.
