Hi,

I have been trying to solve one question in particular after our enzyme kinetics practical but I cannot seem to go any further. Would be able to help me out please. It’s been nearly 4 days now of me trying to solve it in different ways.


Q: Calculate the extinction coefficient (mM^-1.cm^-1) for the 0.5M ethanol (final concentration) sample that displays a maximum absorbance of 2.5. Assume full conversation of NAD+ (100microL in 2.4mL of 20MM solution)?


I appreciate that we have to use Beer-Lambert formula. Absorbance is 2.5, path length is 1.0. I cannot seem to find concentration of NADH using C1V1=C2V2, which will let me find extinction coefficient then.
