Nalidixic acid selectively and reversibly blocks DNA replication in bacteria by inhibiting a subunit of DNA gyrase and topoisomerase IV and suppressing the nicking-closing activity of the DNA gyrase. As a result, nalidixic acid inhibits the release of the positive supercoiling of bacterial DNA.
