Acyclovir (ACV) is converted to its triphosphate form, acyclovir triphosphate (ACV-TP), which competitively inhibits viral DNA polymerase.

It does this by incorporating into the viral DNA chain and inactivates the viral DNA polymerase thereby terminating the growing viral DNA chain.
