I am litter bit confused about how to prepare 25:75 acetonitrile/water ....the problem is that I couldn't get 100 ml as final volume....Do I need to complete the volume to 100 with water?!
So please could you kindly tell me what the correct way to prepare 25:75 acetonitrile/water as media ...Thank you so much for your help

The proportion 25:75 means that 1 part of acetonitrile needs 3 parts of water. For example, if you need to prepare 4 ml of the solution, so you should 1 ml of acetonitrile dilute into the 3 ml of water; for 20 ml solution – 5 ml of of acetonitrile dilute into the 15 ml of water and etc. Thus, for your case you need to take 25 ml of acetonitrile and dilute into the 75 ml of water, general volume is 25 + 75 = 100 ml.
