Chloroquine is a 4-aminoquinoline compound that has been used extensively for the treatment and prevention of malaria.


Chloroquine Phosphate is the active component of chloroquine tablet.


All reagents and solvents used were commercially available and used without further purification. Flash chromatography was performed on Kieselgel 60, particle size 0.032–0.063 mm. NMR spectra were obtained on a 400 MHz (1H-NMR) and 100 MHz (13C-NMR) Varian FT-NMR spectrometer using CDCl3 as solvent. Electrospray mass spectra (ESI-MS) were collected on a Thermo Finnigan LCQ instrument. Samples were dissolved in acetonitrile/water (1:1 v/v) containing 1% acetic acid (1 mg/mL) for MS analysis. Based on NMR spectroscopic and HPLC chromatographic analyses, all compounds were at least of 98% purity.
