Surprisingly, the activity of proteolytic enzymein some isolates increased in presence of 0.6 mM PMSF inhibitor (serineprotease inhibitor). Later, you increased concentration to 6 mM, did not see reduction in activity. The same was observed in 0.1% gelatin zymography(activity staining) as well. The enzyme might be degraded by serine proteases present in the extract. However, PMSF inhibits these proteases increasing the extracted protease activity.
