The function of the primase enzyme is to synthesize short RNA primers that are required to initiate synthesis of new DNA strands by the enzyme DNA polymerase.

Synthesis of primer is the critical step to initiate DNA replication. Thus if the gene for primase is mutated then initiation of DNA replication will be affected.
